Nonexhaustive summary of my projects/work since 1999:

1995-1999: Sherbrooke, Canada
Identification and collection of the insects of North America.
Identification and collection of foams and the lichens.
Identification and collection of the vascular plants of North America and Paris.
Studies of the CO2 assimilation in 2 aquatic environments: river vs. Lake.
Studies on the food preference of the small rodents of Lennoxville.
Studies on the flora and fauna of the salt marshes and the marshes.

1999-2003: My search could show that it was possible to deteriorate the lipidic composition of the plants (the pea) by increasing the rate of fatty-acids polyinsaturés without calling neither upon GMO nor with natural mutants. During these 4 years my work consisted in making push pea seedlings in sterile involved medium or oleic acid (18: 1). The total lipids were then extracted first once by TLC and second once by GC or HPLC.


2004-2007 Paris, National university
The diatoms are photosynthetic organizations marines and of fresh water which, not only hold a key role in the food chain, but which is also important from their production of O2. Indeed, 40% of the Oxygen of our planet come from the oceans and the water bodies, including 20 to 25% directly of the diatoms. With an aim of better including/understanding the mechanisms of assimilation of the nitrogen (NR), I wanted to study a diatom more closely: Phaeodactylum tricornutum whose genome was sequence. My research project consists in locating in this diatom a key enzyme in the metabolism of nitrogen: the nitrate réductase. This enzyme is not only important in the assimilation of nitrogen but also in the assimilation of CO2 (by the production of the amino-acids and proteins). I have clone several transgéniques and studied the localization of several proteins. My search could show that the NR was at the level of the cytoplasm, that this enzyme is sensitive to the source of nitrogen (NO3 vs. NH4) and that this enzyme is mobile in the cell; according to the age of the culture and the luminosity.


The various techniques which I learned and which I was brought to use are the following ones:
Extraction of ARN and DNA, cDNA AFLP, microscopy with fluorescence, microscopy confocale, electronic microscopy, preparation of cuts for electronic microscopy. I was also brought to transform cells by using an embarrassment gun, like making cloning in various stocks of E.coli by using the Gateway system.
